UJI AKTIVITAS ANTIOKSIDAN, KANDUNGAN FENOLIK TOTAL, DAN KANDUNGAN FLAVONOID TOTAL BUAH MERAH (PANDANUS CONOIDEUS LAM.)
Abstrak

Antioksidan merupakan senyawa  yang mampu menghambat reaksi radikal bebas dalam tubuh. Penelitian ini bertujuan untuk menentukan aktivitas antioksidan ekstrak dan fraksi dari buah merah, menentukan kandungan fenolik dan flavonoid total, serta menentukan hubungan antara aktivitas antioksidan dengan kandungan fenolik total dan flavonoid totalnya. Buah merah dimaserasi dengan metanol lalu diuapkan hingga diperoleh ekstrak metanol. Ekstrak metanol dilarutkan dalam air lalu dipartisi dengan kloroform, dan etil asetat untuk memperoleh fraksi kloroform, etil asetat, dan fraksi air. Aktivitas antioksidan ditentukan dengan uji penangkapan radikal DPPH (2,2-difenill-1- pikrilhidrazil), sementara kandungan fenolik total dan flavonoid total ditentukan dengan teknik spektrofotometri. Hasil penelitian menunjukkan bahwa nilai IC50 fraksi air, fraksi etil asetat, fraksi kloroform, dan ekstrak metanol sebelum dipartisi buah merah masing-masing sebesar 399,06; 10,41; 1416,68 dan 186,31 μg/mL. Aktivitas antioksidan ekstrak dan fraksi ini lebih kecil daripada vitamin E (IC50 8,27 µg/ml). Kandungan fenolik total ekstrak dan fraksi buah merah dan nilai IC50-nya menunjukkan hubungan linier y = -13,085x + 754,91; dengan nilai koefisien korelasi (r2) = r2 = 0,3641; sementara itu hubungan flavonoid total ekstrak dan fraksi buah merah dengan IC50-nya mempunyai persamaan regresi linier y = -282,9x + 868,05; dengan nilai  r2 = 0,4484.

PENGARUH PENGGUNAAN ISOFLURAN TERHADAP PERUBAHAN KADAR MALONDIALDEHYDE DAN RASIO EKSTRAKSI OKSIGEN PADA PASIEN PASKA KRANIOTOMI
Abstrak

Traumatic brain injury can increase the production of free radical which is shown by the increasing of MDA content and cell death shown by the ability of oxygen usage by the cell, in this case by observing the oxygen extraction ratio. Isoflurane as neuroprotector can prevent the effect of  free radical. This research aim to get some anesthesi inhalation agent that good enough for neuro anesthesi in brain protection, in term of the decreasing MDA or free radical content and  the oxygen extraction ratio during pre operative, durante operative and post operative. The subject of this research are 19 patients which had craniotomy operation at DR. Sardjito hospital, elective or emergency. Craniotomy operation had done by used of anesthesi with isoflurane inhalation. Blood sample took from jugularis interna vein MDA testing and vein blood gas analyze (SvO2). Blood gas analyze artery (SaO2) took from radialis artery blood sample. Examination had done during pre operative, durante operative and post operative.

The result showed MDA had decreased as 7,49% at durante operatif than at pre operatif and MDA blood content had decreased as 17,30% at post operative than at durante operatif. Oxygen extraction ratio had increased as 20,43% at durante operatif than at pre operatif and had increased as 23,74% at post operative than at durante operatif. It could be conclude that isoflurane had decreased free radical which shown by decreasing MDA at durante operative and post operative than pre operative and isoflurane had increased oxygen extraction ratio which shown by increasing of oxygen usage by the cell at durante operative and post operative than pre operative on patient trauma brain injury.

Antioxidant activity of white and red cabbage (Brassica oleracea L.var capitata L.) using DPPH
Abstrak

Cabbage (Brassica oleracea L.var capitata L) is one of many vegetables in Indonesia. There are two kinds of cabbage white and red cabbage. Phytochemical screening and antioxidant activity have been carried out of raw, steamed and boiled using DPPH. The result of phytochemical screening of white and red cabbage contains flavonoids and steroid/triterpenoid. The result of DPPH method showed that the IC50 value of raw red cabbage, steamed and boiled respectively were 35.84 ppm, 56.83 ppm, and 80.07 ppm. The IC50 value of raw, steamed and boiled white cabbage respectively were 161.23 ppm, 217.51 and 304.59 ppm. From the results showed that antioxidant activity raw red cabbage is the highest but lower than vitamin C as positive control (IC50 = 7,39 ppm)

Biological activity test of extracts and fractions there of keladi tikus leaves (Typhonium flagelliforme (Lodd.) Blume
Abstrak

Abstract: Typhonium flagelliforme (keladi tikus), from familia Araceae, is widely used as an alternative therapy to treat various cancer. In this study, the leaves of keladi tikus plant were extracted using methanol and then partitioned using n-hexane, ethylacetate, and n-buthanol sequentially. The phytochemical screening, and the biological activity using brine shrimp lethality (BSLT) were carried out in this research. The result of phytochemical screening showed that the leaves contained steroids, flavonoids, and saponins. The results of BSLT method showed that the most active extract was ethylacetate extract (LC50 = 55 µg/mL) and the lowest one was the n-buthanol extract (LC50 = 384 µg/mL). The ethyl acetate extract was tested for cytotoxic activity on T-47D cells using the MTT assay method. The IC50 value was 30 µg/mL. The ethylacetae extract was then fractioned by VLC using gradient elution (DCM, isopropanol, and methanol); the fractions were tested using BSLT. The results from the nine fractions showed that  the 5th fraction was the most active causing 97% mortality. It can be concluded that the ethyacetate extract is potential theraupetic bioactive compound.
